چکیده
CONTEXT The identification of genes involved in tumorigenesis is essential for the development of new treatment strategies or diagnostic approaches for pancreatic cancer. OBJECTIVE To identify genes overexpressed in pancreatic cancer we employed differential display, a PCR-based method of differential expression cloning. Using this method, we identified a PCR product that was consistently overexpressed in pancreatic tumors relative to normal pancreatic tissues. SETTING Five pancreatic ductal adenocarcinomas and 5 bulk pancreatic ducts isolated from independent pancreatic specimens without malignancies were analyzed by differential display. A panel of 12 pancreatic tumors at various stages of differentiation and a set of 6 pancreatic ducts without malignancies were then used to verify expression by RT-PCR. RESULTS Sequence analysis of a cDNA detected by differential display revealed that it was a portion of the recently cloned gamma-aminobutyric acid A receptor pi subunit. RT-PCR analysis of a panel of RNAs prepared from pancreatic ducts isolated from organs without malignancies and pancreatic tumors confirmed that that the gamma-butyric acid A receptor pi subunit was significantly overexpressed in pancreatic carcinomas. Analysis of 12 pancreatic tumors revealed that the pi subunit was overexpressed in 10 of the tumors (83%). The expression varied among the tumors, however, overexpression was observed in tumors of each histopathological grade. In contrast, none of the normal pancreatic tissues analyzed displayed high levels of expression. CONCLUSIONS The expression of the GABAA receptor pi subunit may thus play an important role in the pathogenesis of pancreatic cancer.

P145: The Role of γ-Aminobutyric Acid Receptor in The Social Anxiety Disorder
Social anxiety disorder (SAD) is the one of the most common anxiety disorders. Despite its high prevalence, the disorder is still considerably undiagnosed and untreated. The disease places a massive burden on patient’s lives, affecting not only their social interactions but also their educational and professional activities, thereby constituting a severe disability. γ-aminobutyric acid (GABA) s...
متن کاملPrenatal stress increased γ2 GABAA receptor subunit gene expression in hippocampus and potentiated pentylenetetrazol-induced seizure in rats
Objective(s): Stress during pregnancy is able to bring extensive effects on neurobehavioral development in offspring. The GABAergic system plays a pivotal role in neuronal excitability, which can be affected by prenatal stress (PS). This study aimed to evaluate impact of the PS on γ2 subunit of gamma-aminobutyric acid A (GABAA) receptor gene expression in the hippocamp...
متن کاملGABAA Receptor Subunits in Rat Testis and Sperm
Background γ-Aminobutyric acid (GABA) is considered to be the predominant inhibitory neurotransmitter in mammalian central nervous systems (CNS). There are two major classes of GABA receptors: GABAARs and GABABRs. The GABAA receptor is derived from various subunits such as alpha1-alpha 6, beta1-beta 3, gamma1-gamma 4, delta, epsilon, pi, and rho1-3. Intensive research has been performed to und...
متن کاملAn autocrine γ-aminobutyric acid signaling system exists in pancreatic β-cell progenitors of fetal and postnatal mice.
Gamma-aminobutyric acid (GABA) is produced and secreted by adult pancreatic β-cells, which also express GABA receptors mediating autocrine signaling and regulating β-cell proliferation. However, whether the autocrine GABA signaling involves in β-cell progenitor development or maturation remains uncertain. By means of immunohistochemistry we analyzed the expression profiles of the GABA synthesiz...
متن کاملRegulation of Uterine γ-Aminobutyric AcidA Receptor Subunit Expression throughout Pregnancy.
Uterine contractions at parturition depend upon a variety of factors, including gamma-aminobutyric acid (GABA)-ergic stimulation. A new subunit of the GABA(A) receptor, pi, has recently been identified as being particularly abundant in the rat uterus. Reduced derivatives of progesterone, such as the 3alpha,5alpha-reduced derivative termed allopregnanolone, modulate GABA(A) receptor activity and...
